You’ve identified the sequence of a new protein and believe it may be a transmembrane protein. Based on the amino?
acid sequence, you do an analysis to draw a hydropathy plot. What is the basis for the values in the plot? What features of the plot would a probable transmembrane segments in the amino acid sequence have? On the graph below, draw a plot of a 200 amino acid protein that appears to have 2 transmembrane domains.

A hydropathy plot calculates the relative hydrophobicity of continuous small segments (usually 5 amino acids at a time) of the primary structure. A transmembrane segment is usually a stretch of hydrophobic amino acids approximately 20 or so amino acids long.
The graph will have two regions (about 20 aa long) of high hydropathy preceded, separated and followed by segments of low hydropathy.
